To investigate the association of the polymorphisms -866G/A and 518C/T of uncoupling protein 2 (UCP2) gene with metabolic syndrome (MS) and its components in Chinese male. We analyzed the difference of the genetic distribution of UCP2 polymorphism between 496 MS cases and 456 healthy subjects. The differences of genotype and allele frequency distribution for the UCP2-518C/T polymorphism and the UCP2-866G/A polymorphism between MS cases and healthy subjects were examined by logistic regression analysis with adjustment for BMI, age and drinking. The adjusted risk of MS was about 1.8-fold for the UCP2-866 GA genotype (P = 0.041) and 2.2-fold for the UCP2-866 GA+AA genotype (P = 0.010), compared with the wild-type homozygote genotype. In addition, the risk of MS was 1.6-fold for the homozygote mutant genotype (GA and AA) of the UCP2-518C/T polymorphism (P = 0.018) and 2.6-fold for the homozygote mutant genotype (TT) (P = 0.003) compared with the wild-type homozygote genotype (CC). The risks of MS associated with the polymorphism of UCP2-518C/T and UCP2-866G/A were similar in each of the four sex subgroups, and increased gradually with the number of the risk alleles. The genetic polymorphism of UCP2 gene may be associated with metabolic syndrome, suggesting that UCP2 polymorphisms may influence the risk of developing MS.[The assessment of the effectiveness of d0c515b9f4
